Rapid detection of avian leukosis virus subgroup J by cross-priming amplification

چکیده
Abstract Avian leukosis virus subgroup J (ALV-J) causes oncogenic disease in chickens China, resulting great harm to poultry production, and remains widespread China. Herein, we employed a cross-priming amplification (CPA) approach nucleic acid detection device establish visual rapid method for ALV-J. The sensitivity of CPA, polymerase chain reaction (PCR) real-time PCR (RT-PCR) was compared, the three methods were used detect ALV-J cell cultures which inoculated with clinical plasma. result showed when carried out at 60 °C just min, CPA 10 times higher than conventional PCR, high specificity, comparable RT-PCR, based on 123 plasma, coincidence rate 97.3% (71/73). does not require an expensive instrument; simple water bath or incubator is sufficient complete DNA amplification, closed avoids aerosol pollution, making judgment results more intuitive objective. assay would be promising simple, sensitive identification

Avian Leukosis Virus Subgroup J in Layer Chickens, China
To the Editor: In recent years, cases of avian leukosis virus subgroup J (ALV-J) infection and tumors in commercial layer chickens and breeders of egg-type chickens have been emerging in the People’s Republic of China. ALV-J was fi rst isolated from meattype chickens with myeloid leukosis in 1988. Although egg-type chickens have been experimentally infected with ALV-J to induce tumors (1), fi e...
متن کاملMetastatic cutaneous neuro- myofibroblastic sarcoma induced by avian leukosis virus subgroup J in a rooster (Gallus gallus domesticus)
An adult native cock (Gallus gallus domesticus) referred to the aviary clinic with multiple different sizes of round dermal nodules. The bird died few days later, and was then submitted for further evaluation. Macroscopic and microscopic examinations as well as a PCR test were done to identify type and cause of the tumor. In histopathological assessment of biopsy specimen, it consisted of inter...
متن کاملMicroRNA-23b Promotes Avian Leukosis Virus Subgroup J (ALV-J) Replication by Targeting IRF1
Avian leukosis virus subgroup J (ALV-J) can cause several different leukemia-like proliferative diseases in the hemopoietic system of chickens. Here, we investigated the transcriptome profiles and miRNA expression profiles of ALV-J-infected and uninfected chicken spleens to identify the genes and miRNAs related to ALV-J invasion. In total, 252 genes and 167 miRNAs were differentially expressed ...
متن کاملDetection and Molecular Characterization of J Subgroup Avian Leukosis Virus in Wild Ducks in China
To assess the status of avian leukosis virus subgroup J (ALV-J) in wild ducks in China, we examined samples from 528 wild ducks, representing 17 species, which were collected in China over the past 3 years. Virus isolation and PCR showed that 7 ALV-J strains were isolated from wild ducks. The env genes and the 3'UTRs from these isolates were cloned and sequenced. The env genes of all 7 wild duc...
